Answered: Asking about Deuterium NMR
I wanted to do Deuterium NMR in human urine sample but no one has done previously in our lab.
In our Lab. we have 400 MHz Bruker (Avance) spectrometer with BroadBand Inverse probe (1H Observe, Z-gradient).
How we have to proceed without deuterium lock which is neccessory for exp. ?
Is above described instruments specification sufficient to perform the deutrium NMR experiments ?
Please any on can help

Do you have your human urine sample labeled with deuterium !!! Just kidding.... All you have to do is switch off the lock using command "locnuc=off" and use the sequence ZG2H (read the sequence carefully). Well, this is a general instruction to run 2H spectrum in a Bruker spectrometer. If you fry the probe without taking proper care, i am not responsible for it !!!

An alternative method to switching off the D-Lock is the use of capilar tubes, I use a NewEra's capilar tube for 500 µL of sample and 60 µL of D2O in the capilar

If I'm running relaxation experiments (15N) for R1, R2, and hetnoe on a double labeled (2H/15N) sample, do I need to do any deuterium decoupling added to the program?

Hello, I am trying to investigate what has been done in NMR using direct detection of deuterium in liquids. - Could you suggest any citations?
What kind of sensitivity gain there is in using broadband probe vs. lock channel?
Has anything been done/can be done to detect scalar couplings with protons like in COSY or TOCSY? Are HD couplings other than geminal detectable?
