Hello, I am trying to investigate what has been done in NMR using direct detection of deuterium in liquids. - Could you suggest any citations?
What kind of sensitivity gain there is in using broadband probe vs. lock channel?
Has anything been done/can be done to detect scalar couplings with protons like in COSY or TOCSY? Are HD couplings other than geminal detectable?
Thanks.

Speeding up direct 15N detection: hCaN 2D NMR experiment
Abstract Experiments detecting low gyromagnetic nuclei have recently been proposed to utilize the relatively slow relaxation properties of these nuclei in comparison to 1H. Here we present a new type of 15N direct-detection experiment. Like the previously proposed CaN experiment (Takeuchi et al. in J Biomol NMR 47:271â??282, 2010), the hCaN experiment described here sequentially connects amide 15N resonances, but utilizes the initial high polarization and the faster recovery of the 1H nucleus to shorten the recycling delay. This...

[Question from NMRWiki Q&A forum] How to set up 15N direct detection on UnityInova?
How to set up 15N direct detection on UnityInova?
Has anyone recorded directly detected 15N spectra on UnityInova? We have a triple resonance HCN probe. The sample I've used was natural abundance 1M NaNO3 - sodium nitrate.
I've tried to set up cable analogously to 13C detection - through the second preamp, but could not see any signal at all.

CACA-TOCSY with alternate 13Câ??12C labeling: a 13Cα direct detection experiment for
Abstract We present a 13C direct detection CACA-TOCSY experiment for samples with alternate 13Câ??12C labeling. It provides inter-residue correlations between 13Cα resonances of residue i and adjacent Cαs at positions i â?? 1 and i + 1. Furthermore, longer mixing times yield correlations to Cα nuclei separated by more than one residue. The experiment also provides Cα-to-sidechain correlations, some amino acid type identifications and estimates for Ï? dihedral angles. The power of the experiment derives from the alternate 13Câ??12C labeling with glycerol or glycerol, which allows...

Asking about Deuterium NMR
I wanted to do Deuterium NMR in human urine sample but no one has done previously in our lab.
In our Lab. we have 400 MHz Bruker (Avance) spectrometer with BroadBand Inverse probe (1H Observe, Z-gradient).
How we have to proceed without deuterium lock which is neccessory for exp. ?
Is above described instruments specification sufficient to perform the deutrium NMR experiments ?
Please any on can help
Thanks very much
