Hi everybody, This is the first post I am sending to this group, I hope you could help me with my question. I will be grateful. I have assigned the NOEs in

Hi,
I have observed some interesting NOEs for my peptide (5 kDa) in my 2D NOESY spectrum recorded at 900 MHz. I see NOEs that forms an α-helical pattern between 12-24 residues and N&C-terminus of my peptide is unstructured and does not have any secondary structure. The interesting part is, apart from seeing helical NOE patterns, such as αN(i,i+3)and αβ(i,i+3) for the residues between 12 & 24, I see in the alpha proton chemical shift regions of my 2D NOESY spectrum, a continuous αH(i,i+1) NOEs for the residues where I have helical secondary structural region (12-24 aa). That is, I see NOE...

Reduced dimensionality 3D HNCAN for unambiguous HN, CA and N assignment in proteins

We present here an improvisation of HNN (Panchal, Bhavesh et al., 2001) called RD 3D HNCAN for backbone (HN, CA and 15N) assignment in both folded and unfolded proteins. This is a reduced dimensionality experiment which employs CA chemical shifts to improve dispersion. Distinct positive and negative peak patterns of various triplet segments...

[Question from NMRWiki Q&A forum] Varian Biopack pulse sequences for intermolecular NOEs
Varian Biopack pulse sequences for intermolecular NOEs
Hi All, I'm trying to set up some experiments to determine intermolecular NOEs in a peptide:protein complex. At the moment from looking at the Biopack library, I have found 4 pulse sequences which will do this and each provide different information (gnoesyNhsqc_CN, gnoesyNhsqc_NN, gnoesyChsqc_CC and gnoesyChsqc_NC). Would people recommend using a combination of all four of these experiments to determine the intermolecular NOEs for use in structure calculations?

The completeness of experimentally observed NOE restraints of a set of 97 NMR protein structures deposited in the PDB has been assessed. Completeness is defined as the ratio of the number of experimentally observed NOEs and the number of 'expected NOEs'. A practical definition of 'expected...
