I have not fully understood the mechanism of the filtered NOE experiment. What does it tell you? Are you able to distinguish between inter- and intradomains in a protein? How is such an experiment performed in the lab?

[Question from NMRWiki Q&A forum] Isotope Filtered NOESY
Isotope Filtered NOESY
I'm an NMR newbie. I know isotope filtering can be used to study a complex under slow exchange, but for fast-exchange, is it possible to acquire only the intramolecular NOEs of the receptor as opposed to intermolecular NOEs between the receptor and ligand? I just want the contacts in the receptor. Thanks.

[Question from NMRWiki Q&A forum] 2D Filtered NOESY/TOCSY on Varian Biopack
2D Filtered NOESY/TOCSY on Varian Biopack
I am attempting to run some filtered NOESY and TOCSY experiments using sequences in the Varian Biopack - CNfiltocsy and CNfilnoesy. The experiments seem to run fine, but when overlaying the processed spectra something seems odd - the TOCSY crosspeaks are not present in the NOESY spectra and vice-versa. Does anyone know why this might be the case?

A double-editing pulse sequence has been developed that allows the direct observation of protein binding ligand(s) from a mixture of compounds. This technique should aid the discovery of lead pharmaceutical compounds. The proton NMR signals from protein and the nonbinding ligands are simultaneously eliminated using 13C isotope editing and PFG diffusion-edited NMR. This new...
