Highly selective and efficient water signal suppression is indispensable in biomolecular 2D nuclear Overhauser effect spectroscopy (NOESY) experiments. However, the application of conventional water suppression schemes can cause a significant or complete loss of the biomolecular resonances at and around the water chemical shift (Ï?2). In this study, a new sequence, NOESY-WaterControl, was developed to address this issue. The new sequence was tested on lysozyme and bovine pancreatic trypsin inhibitor (BPTI), demonstrating its efficiency in both water suppression and, more excitingly, preserving water-proximate biomolecular resonances in Ï?2. The 2D NOESY maps obtained using the new sequence thus provide more information than the maps obtained with conventional water suppression, thereby lessening the number of experiments needed to complete resonance assignments of biomolecules. The 2D NOESY-WaterControl map of BPTI showed strong bound water and exchangeable proton signals in Ï?1 but these signals were absent in Ï?2, indicating the possibility of using the new sequence to discriminate bound water and exchangeable proton resonances from non-labile proton resonances with similar chemical shifts to water.

1D NOESY with selective excitation and water suppression
Hi,
I am trying to collect 1D NOESY spectrum of a small protein in 90% H2O with selective excitation so I could follow a few NOEs without running a whole 2D experiment. Is there any sequence in Varian BioPack that I could use for that? I cannot achieve water suppression with the standard 'Noesy1D' sequence and all the other seem to not allow to selectively irradiate only one peak. Maybe it is just a matter of right parameters but I don't know how to set them properly - if so I will appreciate any advice.

What is best pulse sequence in bio pack on vnmrj for C-13 edit NOESY ? I am working with 12 kd size protein which can exist as homo dimers ( i.e total size of homo-dimer is 24 kd ). Most frequent problems we are encountering as follows !line broadening , baseline , phase correction problems ( huge signal at 2.8 ppm )in first increments and f2f3 planes huge strips errors !

I got doubt that can I use Randomly duterated protein (40 to 60 percent ) to get 3D (C13 EDIT and N15 EDIT) noesy experiments ? because if i duteriated the protein randomly the peak intensity drop would be there. but from standard formula NOE = 1/R6.
I am afraid of error from intensity drop .
could you please suggest any reference related to this ?
