NMR has played an important role in determining the structures, dynamics and interactions of a wide range of plant proteins. This article provides an overview of the NMR techniques that have been used in these studies and describes selected examples from major classes of plant proteins to illustrate applications of these techniques. These classes include defensins, cyclotides, lectins, photosynthetic proteins, transcription factors, and storage proteins. The NMR studies have provided insights into the binding interactions and/or modes of action of many of these proteins and have been instrumental in the development of structure-activity relationships. In addition to just being the subjects of study, plant proteins have played a prominent role in the development of many modern NMR techniques. This role has been driven by the ready availability of a number of important ‘model’ proteins from plants and also by the generally high stability of plant proteins. For example, some of the early developments of multidimensional NMR techniques relied heavily on the availability of large quantities of stable plant proteins, e.g., crambin, protease inhibitors and several metallo-proteins. Overall, plant proteins have been valuable subjects and tools for NMR analysis.
