Default Development of PRC1 Inhibitors Employing Fragment-Based Approach and NMR-Guided Optimization

Development of PRC1 Inhibitors Employing Fragment-Based Approach and NMR-Guided Optimization

Polycomb Repressive Complex 1 (PRC1) is associated with transcriptional silencing, and its dysregulation plays an important role in various cancers. Well-characterized PRC1 inhibitors can facilitate the exploration of PRC1 inhibition as therapeutic agents. By employing an NMR-based fragment screening approach, we have previously identified a very weak millimolar ligand RB-1, which directly binds to RING1B-BMI1. Then, we reported a low-micromolar PRC1 inhibitor, RB-3, which is active in leukemic...

[NMR paper] Investigate the metabolic changes in intestinal diseases by employing a (1)H-NMR-based metabolomics approach on Caco-2 cells treated with cedrol
Investigate the metabolic changes in intestinal diseases by employing a (1)H-NMR-based metabolomics approach on Caco-2 cells treated with cedrol Mitochondrial dysfunction may precipitate intestinal dysfunction, while inflammatory bowel disease manifests as a chronic inflammatory ailment affecting the gastrointestinal tract. This condition disrupts the barrier function of the intestinal epithelium and alters metabolic products. Increasing mitochondrial adenosine triphosphate (ATP) synthesis in intestinal epithelial cells presents a promising avenue for colitis treatments. Nevertheless, the...

[NMR paper] A Step Towards NRF2-DNA Interaction Inhibitors by Fragment-Based NMR Methods
A Step Towards NRF2-DNA Interaction Inhibitors by Fragment-Based NMR Methods The NRF2 transcription factor is a key regulator in cellular oxidative stress response, and acts as a tumor suppressor. Aberrant activation of NRF2 has been implicated in promoting chemo-resistance, tumor growth, and metastasis by activating its downstream target genes. Hence, inhibition of NRF2 promises to be an attractive therapeutic strategy to suppress cell proliferation and enhance cell apoptosis in cancer. Direct targeting of NRF2 with small-molecules to discover protein-DNA inhibitors is... More...
