Default Determination of Amide Cis/Trans Isomers in N-Acetyl-d-glucosamine: Tailored NMR Analysis of the N-Acetyl Group Conformation

Determination of Amide Cis/Trans Isomers in N-Acetyl-d-glucosamine: Tailored NMR Analysis of the N-Acetyl Group Conformation

N -Acetyl- d -glucosamine (GlcNAc) is one of the most common amino sugars in nature, but the conformation of its N -acetyl group has drawn little attention. We report herein the first identification of NH protons of the amide cis forms of ? - and ? -GlcNAc by NMR spectroscopy. Relative quantification and thermodynamic analysis of both cis and trans forms was carried out in aqueous solution. The NH protons were further utilized by adapting protein NMR experiments to measure eight J -couplings...

[NMR paper] An integrative NMR-SAXS approach for structural determination of large RNAs defines the substrate-free state of a trans-cleaving Neurospora Varkud Satellite ribozyme
An integrative NMR-SAXS approach for structural determination of large RNAs defines the substrate-free state of a trans-cleaving Neurospora Varkud Satellite ribozyme The divide-and-conquer strategy is commonly used for protein structure determination, but its applications to high-resolution structure determination of RNAs have been limited. Here, we introduce an integrative approach based on the divide-and-conquer strategy that was undertaken to determine the solution structure of an RNA model system, the Neurospora VS ribozyme. NMR and SAXS studies were conducted on a minimal trans VS...

[NMR paper] Selective 1Halpha NMR methods to reveal functionally relevant proline cis/trans isomers in IDPs: characterization of minor forms, effects of phosphorylation and occurrence in proteome
Selective 1Halpha NMR methods to reveal functionally relevant proline cis/trans isomers in IDPs: characterization of minor forms, effects of phosphorylation and occurrence in proteome Identification of proline cis/trans isomers appearing in several regulatory mechanisms of proteins and characterization of minor species present due to the conformational heterogeneity in IDPs is highly important. To obtain residue level information on these mobile systems we introduce two 1 H ? -detected, proline selective, real-time homodecoupled NMR experiments and analyze the proline abundant...

Observation of acetyl phosphate formation in mammalian mitochondria using real-time in-organelle NMR metabolomics [Biochemistry]
Observation of acetyl phosphate formation in mammalian mitochondria using real-time in-organelle NMR metabolomics Wen Jun Xu, He Wen, Han Sun Kim, Yoon-Joo Ko, Seung-Mo Dong, In-Sun Park, Jong In Yook, Sunghyouk Park... Date: 2018-04-17 Recent studies point out the link between altered mitochondrial metabolism and cancer, and detailed understanding of mitochondrial metabolism requires real-time detection of its metabolites. Employing heteronuclear 2D NMR spectroscopy and 13C3-pyruvate, we propose in-organelle metabolomics that allows for the monitoring of mitochondrial metabolic...

NMR profiling of histone deacetylase and acetyl-transferase activities in real time. NMR profiling of histone deacetylase and acetyl-transferase activities in real time. ACS Chem Biol. 2011 May 20;6(5):419-24 Authors: Dose A, Liokatis S, Theillet FX, Selenko P, Schwarzer D Abstract Histone deacetylases (HDACs) and histone acetyl-transferases (HATs) are universal regulators of eukaryotic transcriptional activity and emerging therapeutic targets for human diseases. Here we describe the generation of isotope-labeled deacetylation and...
